Teddy (Zac Efron) is living with frat brother Pete (Dave Franco), and it is revealed that Pete has come out of the closet since college. Pete asks him to move out after his boyfriend proposes to him. Three college freshmen - Shelby (Chloë Grace Moretz), Beth (Kiersey Clemons) and Nora (Beanie Feldstein) - are rushing for a sorority. After discovering that official sororities can't throw their own parties and fraternity parties are disgusting and misogynistic, the trio decide to found their own sorority off-campus in order to be afforded more freedom than what is offered by traditional sororities.

The three girls come across the former fraternity house in which Teddy and Pete lived that is located next door to Mac and Kelly's house. They discover Teddy wallowing in sorrow inside. He convinces the girls to rent his former frat house and let him live there as their advisor. Mac and Kelly try to get the girls to keep things quiet until escrow closes, but the sorority and Teddy refuse to cooperate and a prank war ensues. Mac and Kelly's married friends Jimmy (Ike Barinholtz) and Paula (Carla Gallo) also pitch in to help Mac and Kelly.

In need of rent money, the sorority girls decide to sell marijuana. Teddy tries to convince them out of this plan, so they fire him as advisor and kick him out of the house. He approaches Mac and Kelly and aligns with them to get revenge on the sorority. Mac, Kelly, Jimmy, Paula and Teddy enact a plan to successfully steal the sorority's weed before they can sell it. They believe the loss of the weed will force the girls to miss their rent payment and be evicted.

Realizing they have no other choice, the girls decide to throw a frat-style party and charge entrance fees to get the money necessary to cover rent. he girls dress in provocative clothing and attract many men from college. They also successfully stop Mac, Kelly and Teddy from shutting the party down. During the party, the buyers discover the sorority living and withdraw their offer to buy Mac and Kelly's home. Although the girls appear victorious, they realize that they have betrayed the values of their sorority and have a falling out. Realizing that salvaging the girls' self-respect and confidence is more important than their feud, Mac, Kelly and Teddy convince the girls that they can succeed on their terms. The girls kick out all the misogynistic college boys from their party and hold the kind of party they envisioned when the started the sorority. As a result, every girl on campus decides to join their sorority and pay dues to them.

That said, the girls missed the deadline to pay their rent payment and are being evicted. Now on good terms with each other, the sorority reach a deal to rent Mac and Kelly's home (thus allowing Mac, Kelly and their family to afford to move to their new home).

In the end, Mac and Kelly are peacefully living in the suburbs and going on play-dates with Paula, Jimmy and their child. Teddy has discovered that his skills for devising and implementing complex plans during the prank wars makes him perfectly suited to work as a wedding planner (thus, he finally moves on from his college mindset). He also is the Best Man at Pete's wedding.
